Dive Objectives Brief:

Primary Objective — Arrive at P-vent to slurp and Collect ASU Arnold, move to Bio-9 and collect Frying Pan, Take Sippers of each, Alvinella collection at Bio-9, collect Harold samplers from LVWS site, move Harold sampler into LVWS site from Neo-Fish Hole, collect Mosquito with Sipper, move to TICA — deploy Cavanaugh samplers, collect small Riftia in tubes and medium worm, cut open into Shank Sampler, pick up nice piece of chimney with chalcopyrite (gold) for ship-wide distribution.

Sampling procedure:

FOR ALL SIPPER SAMPLES — DON’T FORGET TO TURN PUMP ON!!!!!!!!

Alvinella:

1) Take temp. of several tubes with Sipper wand — if over 50°C take Sipper chem.

2) Collect worms — place into Biobox or DISSR.

3) Take temp. and chem. of bare sulfide surface where tubes were collected.

Beehive/chimney:

1) Take temp. and Sipper chem. of beehive on side and top.

2) If feature has orifice, do not put Sipper in it — take temp./chem. on side.

3) Collect beehive and place in lg. Biobox.

ASU Recovery (Frying pan, foreplay, Arnold):

1) Take temp. at unit.

2) Take Sipper/chem.

3) Remove ASU unit from sulfide.

4) Place in coffin.

Dive Objectives:

1) While descending, check SIPPER, temp. probe for function.

2) Move into P-vent.

3) Locate Arnold at P-vent. (X=4666 Y=77914, heading = 024).

4) Get temp. and take Sippers — one native, one charged.

5) Remove and place into far left compartment of Biobox.

6) Get temp. and take Sipper of scar — one of each.

7) Go to top of P-vent and slurp white flock on chimney — 10 min. only.

8) Get temp.

9) Move to Bio-9.

10) Move to the Frying Pan (X = 4661, Y = 77958, Heading = 327).

11) Sample the same as for the Arnold.

12) Place Arnold unit into middle compartment of large Biobox.

13) Move in to do Alvinella sampling.

14) Locate chimney with hot Alvinella — this can be at Bio-9/P-vent/Alvinella Stump if necessary.

15) Position for temp. survey — use Sipper Probe if working — if not use Hi-T.

16) Take Sipper sample/temp. from inside tubes — X 3–6. Get good visuals (video and still) if temps. are above 50°C.

17) Collect large clump of hot (65°C) Alvinella. Place into right-most chamber of lg. Biobox with ice. Try not to collect sulfides with worms. Use camera to get idea of number and size.

18)  Take your time here — may be best to grab clump and shake above DISSR.

19) Collect Alvinella and place onto coffin lid — sort for DISSR.

20) Collect Alvinella and place into DISSR — close lid.

21)  Move in to collect Harold samplers at LVWS site (X = 4653, Y = 77953, heading = 322).

22) Get temp. of bacteria-covered unit.

23) Take Sipper.

24)  Place both in small Cary Biobox.

25) Move Harold sampler in Neo-Fish Hole to LVWS site — get X,Y and headings.

26) Collect Mosquito.

27) Go to TICA.

28) Place 2 Cavanaugh collectors at 10’ intervals in line from clump.

29) Place last unit 90° to right or left of fourth unit — get (X= Y=, and headings).

30) Look for small Riftia at base for collection tomorrow.

31) Place a small clump into Shank unit.

32) Grab a medium worm. De-tube and cut 2/3 down.

33) Let bleed a moment — place into Shank sampler.

34) Put in Shank stopper — FIRE.

35) On ascent — please place manipulators on coffins if possible.

Detailed instructions:

1) On descent, review still and video (hand-held and sub) camera operation.

2) Review Sipper/AMS operations.

3) Land EAST of ASC at latitude of Q-vent.

A. Collect ASU Arnold Unit.

1) Move into P-vent (X = 4666 Y = 77914, heading = 024)

2) Locate Arnold unit and prepare to recover.

3) Get good video with 3-chip and no overlay.

4) Take Sipper temp. and chem. (X 2 one charged, one uncharged).

5) Recover — be very careful.

6) Place in Biobox (left or middle section in lg. box).

7) Take 2 Sipper temp. and chem. (charged and uncharged) after removal.

B. Slurp white flock on P-vent chimney.

 1) Move over to spire at P-vent with white flock.

 2) Use slurp to suck up white flock on chimney — spend no more than 10 mins.

Collection of Alvinella can be made at P-vent or Bio-9.

C. ALVINELLA

We are going to do chem. and temp work on Alvinella — but we really want Hot guys (lots of shimmering water). Please take lots of pics of each procedure.

This can be done at Bio-9 or at P-vent.

a.  SURVEY

 1) Set up and do some preliminary temp. checks to make sure temps. are high (>50°C) at high-temp. site.

1) Take hand-held video/still of Alvinella before sampling. Position 3-chip to get some good video of the worms moving in and out of tube and sample area.

2) Take internal temp. of several tubes; record on dive sheet. The avg. temp inside tubes should be 65°C (45°C outside) or greater.

3) Use the Sipper temp. probe and record data from overlays on data sheets.

4) If hot, take temps. in tubes(X3 tubes) and Sipper x3 (different tubes). Draw picture of positions — note time and location on data sheet.

b. Alvinella — DISSR

IN ALL CASES WE NEED TO HAVE TEMP. AND SIPPER DATA FROM EACH COLONY SAMPLED.

5) Position overhead camera so that it is looking down into DISSR Sampler. The other camera should be recording Alvinella clump.

6)  Move a clump to the top of the coffin — sort and drop animals into DISSR. Look to make sure you have worms — 10 – 20.

7) Collect 4–6 large (6–8 med.) worms without damaging them — avoid tube and debris if possible — they take time to sink. Make sure they go down.

8) Need good visuals of worms going into sampler — note time.

9) Take good still shot/video of worm going into box for Ambion — NOTE.

c. COLLECT MASS ALVINELLA CLUMP.

10) Collect lg. Alvinella clump and place in single chamber of Biobox with ice (RIGHT).

11) See if you can get a good clump without sulfide.

12) Visualize number collected.

13) Repeat procedure until you have a good number of worms in box (20–30).

14) Take Final Sipper and Temp. on the surface of the sulfide from where the worms were sampled — note on dive summary report sheet XY and headings.

MOVE TO BIO-9

D. Collect ASU Frying Pan Unit.

8) Move into P-vent (X = 4661 Y = 77958, heading = 327).

9) Locate Frying Pan unit and prepare to recover.

10) Get good video with 3-chip and no overlay.

11) Take Sipper temp. and chem. (X2 one charged, one uncharged).

12) Recover — be very careful.

13) Place in Biobox (left or middle section in lg. box).

14) Take 2 Sipper temp. and chem. (charged and uncharged) after removal.

E. Collect Mosquito Probe

1) Take sipper/chem. in same hole as Mosquito.


F. COLLECT 2 HAROLD SAMPLERS

1) Move into Bio-9 LVWS site — (X = 4653, Y = 77953, heading= 322).

2) Locate Harold samplers.

3) Take one Sipper on sampler with most bacteria.

4) VERY GENTLY collect and place in small Cary Biobox.

G. MOVE HAROLD SAMPLER IN NEO-FISH HOLE.

1) Move sampler to LVWS site — get good X,Y and heading.

Move to TICA

H. Deploy 3 Cavanaugh samplers.

1) Place one 10’ down strike from unit placed there yesterday.

2) Get XY and heading.

3) Place 4th 5–10 feet away down ASC.

4) Get XY and heading.

5) Place 5th unit 90° to East or West of 4th unit.

6) Get XY and heading.

I . Move into East wall at base to collect small Riftia.

1) Find a small clump of small Riftia.

2) Place into Shank sampler.

3) Collect one medium worm — cut 2/3 down and let bleed.

4) Place into Shank sampler — cap and fire.

J. If time permits, COLLECT CHALCOPYRITE SAMPLE OF CHIMNEY.
